چکیده

In this paper, we experimentally studied both the bright-field and fluorescence images of microspheres by conventional and confocal scanning polarization microscopes. A qualitative analysis have been given to show a physical picture on the imaging of the microspheres. Emission spectra from melamine formaldehyde microspheres stained with Ethidium Bromide or covered by thin shell of CdTe nanocrystals have been experimentally studied. We adopted analytical expressions describing the resonance spacing in order to determine the size of the microspheres.

Dual-detection confocal fluorescence microscopy: fluorescence axial imaging without axial scanning.

We propose a new method for high-speed, three-dimensional (3-D) fluorescence imaging, which we refer to as dual-detection confocal fluorescence microscopy (DDCFM). In contrast to conventional beam-scanning confocal fluorescence microscopy, where the focal spot must be scanned either optically or mechanically over a sample volume to reconstruct a 3-D image, DDCFM can obtain the depth of a fluore...

A new method for imaging and 3D reconstruction of mammalian cochlea by fluorescent confocal microscopy.

Traditional methods for anatomical and morphometric studies of cochlear tissues have relied upon either microdissection of the organ of Corti or the generation of serial sections of the cochlea. Such methods are time-consuming, disruptive to three-dimensional relationships and often restrict sampling to very limited numbers of cells. We have found that cells and tissue components of the cochlea...

Fast 100-nm resolution three-dimensional microscope reveals structural plasticity of mitochondria in live yeast.

By introducing beam-scanning multifocal multiphoton 4Pi-confocal microscopy, we have attained fast fluorescence imaging of live cells with axial super resolution. Rapid scanning of up to 64 pairs of interfering high-angle fields and subsequent confocal detection enabled us to perform three to five times finer optical sectioning than confocal microscopy. In conjunction with nonlinear image resto...

Variations of the Normal Human Limbal Stem Cells Detected by In Vivo Confocal Microscopy

Background  To report normal variations of the limbal structures using in vivo laser scanning confocal microscopy. Methods: This was a retrospective study of fourteen eyes from 11 healthy individuals. Confocal imaging of cornea and limbus was performed using Heidelberg Retina Tomograph III Rostock Corneal Module. Results: The typical structure of the palisades of Vogt (POV) was detected ...

Three-dimensional imaging of the intact mouse cochlea by fluorescent laser scanning confocal microscopy.

The complex anatomy of the mammalian cochlea is most readily understood by representation in three-dimensions. However, the cochlea is often sectioned to minimize the effects of its anatomic complexity and optical properties on image acquisition by light microscopy. We have found that optical aberrations present in the decalcified cochlea can be greatly reduced by dehydration through graded eth...
